Teen concussions increase risk for depression

Jan. 9, 2014 — Teens with a history of concussions are more than three times as likely to suffer from depression as teens who have never had a concussion, finds a new study in the Journal of Adolescent Health.

"What this study suggests is that teens who have had a concussion should be screened for depression," said lead study author Sara Chrisman, M.D., a pediatrician at Seattle Children's Hospital.

Concussion, considered a mild traumatic brain injury, can also have serious psychological effects. Most prior research on these psychological effects has focused on adults. However, many teens experience concussions through sports injuries or accidents, and less is known about long-term complications in adolescents.

The study used data from the 2007-2008 National Survey of Children's Health and included health information from over 36,000 adolescents ages 12 to 17. 2.7 percent of the sample had had a concussion and 3.4 percent had a current depression diagnosis.

Teens who were 15 years or older, lived in poverty or who had a parent with mental health problems were more likely to be depressed than other teens, said Chrisman, "but what was surprising was when we took those factors into consideration, it didn't take away from the association between depression and a history of concussion."

Chrisman also cautioned that it's not known what exactly might account for higher rates of depression in teens with a history of concussion. It could be the brain injury itself, diagnostic bias due to repeated medical visits for concussion, doctors mistaking symptoms of a concussion for depression, or from the social isolation that they may experience while recovering.

Jeffrey Max, M.D., a psychiatrist who specializes in psychiatric outcomes of traumatic brain injury in children and adolescents at the University of California, San Diego noted, "In our research, we've found that about 10 percent of the kids had a full depressive disorder or subclinical depressive disorder 6 months after a concussion." Children who have a history of concussion are more likely to develop attention-deficit/hyperactivity disorder (ADHD) and have difficulties controlling their moods, especially anger, rather than experience depression, Max added.

Unlike Chrisman, however, Max observed that the actual brain injury associated with concussions is probably a major cause of depression in the first few months after injury. "In the clinic, we've certainly seen cases where within hours [of sustaining a concussion], a kid who's never had depression before is suddenly depressed and suicidal. One of our studies found that the brain images in children with traumatic brain injury and depression were actually quite similar to those seen in adults who develop depression as a result of traumatic brain injury."

Researchers invent 'sideways' approach to 2-D hybrid materials

The study, published in the journal Science, could enable the use of new types of 2-D hybrid materials in technological applications and fundamental research.

By rethinking a traditional method of growing materials, the researchers combined two compounds -- graphene and boron nitride -- into a single layer only one atom thick. Graphene, which consists of carbon atoms arranged in hexagonal, honeycomb-like rings, has attracted waves of attention because of its high strength and electronic properties.

"People call graphene a wonder material that could revolutionize the landscape of nanotechnology and electronics," ORNL's An-Ping Li said. "Indeed, graphene has a lot of potential, but it has limits. To make use of graphene in applications or devices, we need to integrate graphene with other materials."

One method to combine differing materials into heterostructures is epitaxy, in which one material is grown on top of another such that both have the same crystalline structure. To grow the 2-D materials, the ORNL-UT research team directed the growth process horizontally instead of vertically.

The researchers first grew graphene on a copper foil, etched the graphene to create clean edges, and then grew boron nitride through chemical vapor deposition. Instead of conforming to the structure of the copper base layer as in conventional epitaxy, the boron nitride atoms took on the crystallography of the graphene.

"The graphene piece acted as a seed for the epitaxial growth in two-dimensional space, so that the crystallography of the boron nitride is solely determined by the graphene," UT's Gong Gu said.

Not only did the team's technique combine the two materials, it also produced an atomically sharp boundary, a one-dimensional interface, between the two materials. The ability to carefully control this interface, or "heterojunction," is important from an applied and fundamental perspective, says Gu.

"If we want to harness graphene in an application, we have to make use of the interface properties, since as Nobel laureate Herbert Kroemer once said 'the interface is the device,'" Li said. "By creating this clean, coherent, 1-D interface, our technique provides us with the opportunity to fabricate graphene-based devices for real applications."

The new technique also allows researchers to experimentally investigate the scientifically intriguing graphene-boron nitride boundary for the first time.

"There is a vast body of theoretical literature predicting wonderful physical properties of this peculiar boundary, in absence of any experimental validation so far," said Li, who leads an ORNL effort to study atomic-level structure-transport relationships using the lab's unique four-probe scanning tunneling microscopy facility. "Now we have a platform to explore these properties."

The research team anticipates that its method can be applied to other combinations of 2-D materials, assuming that the different crystalline structures are similar enough to match one another.

Although red blood cells were first visualized in the mid-17th century and studied extensively since then, this new study, published online ahead of print in the journal Blood, describes a previously unknown shape and potential new function for red blood cells. The Penn team found that red blood cells can be compressed into many-sided, closely-packed polyhedral structures -- instead of their free-flowing bi-concave, disc shape.

What's more, contrary to expectations, the aggregates of fibrin and platelets that make up highly contracted clots lie primarily on the clot exterior, with the red blood cells crowded within the clot interior, although the contents of clots are more homogeneous before contraction takes place.

Contracted clots may form an impermeable seal and help prevent vascular obstruction, but confer resistance to penetration by drugs that break down fibrin, the structural component of clots, a common treatment option for heart attacks and strokes.

"The first time we saw this, we thought: 'This can't be biological,'" recalls Weisel. The team first saw the polyhedron-shaped red blood cells when studying the clot-contraction process using a novel magnetic resonance technology, with co-authors from T2 Biosystems, along with co-author Douglas Cines, M.D., director of the Coagulation Laboratory and professor of Pathology and Laboratory Medicine at Penn. They observed a signal that indicated tightly packed red blood cells.

The Clot Network Clots are a three-dimensional network of fibers, made up primarily of the blood protein fibrinogen, which is converted to fibrin during clotting, and platelets, which aggregate by binding to fibrin once they are activated. A blood clot needs to have the right degree of stiffness and plasticity to stem the flow of blood when tissue is damaged, yet be flexible enough that it does not block blood flow.

After a clot forms, the actin and myosin in platelets start the contraction process and cause the clot to shrink to about one-third of its original size. This is an important step to stem bleeding, to decrease obstruction in blood vessels, and to provide a matrix for migration of cells involved in wound healing. Red blood cells are caught up in the contraction process, especially in the venous system, and get pulled by the platelets towards the interior of the clot, as the Blood study indicates.

Little was known about the structure of contracted clots or the role of red blood cells in the contraction process. "We found that contracted blood clots develop a remarkable structure, with a meshwork of fibrin and platelet aggregates on the exterior of the clot and a close-packed, tessellated array of compressed polyhedral erythrocytes within," says Weisel.

The team also saw the same morphology of compacted clots after initiating clotting with various activators and also with clots formed from human blood reconstituted with its cellular and plasma components and from mouse blood. Such close-packed arrays of polyhedral erythrocytes, or polyhedrocytes as the researchers dubbed them, were also observed in human arterial thrombi taken from patients who had heart attacks. This shape is likely taken on by the red blood cells when contracted or pushed together when the platelets compress a clot so as to decrease volume, surface energy, or bending energy, surmise the authors.

Cines notes that these findings might have clinical implications. Physicians need to inject thrombolytic agents such as tPA to rapidly break up thrombi, clots that obstruct blood flow, for example in coronary arteries to treat a heart attack, or in the arteries to the brain to treat a stroke. It is well known that with time thrombi develop resistance to being broken up, which is one reason why early intervention is so important. The nearly impermeable barrier formed by red blood cells within the contracted clots that was observed in the Blood study may help to explain why. Clot contraction could be a target of intervention to prevent the formation of the closely packed array of polyhedrocytes.

One species, two outcomes: Team seeks source of body louse pathology

Jan. 10, 2014 — A new study seeks to determine how one parasitic species can give rise to two drastically different outcomes in its host: The human body louse (Pediculus humanus) can transmit dangerous bacterial infections to humans, while the human head louse (also Pediculus humanus) does not.

A report of the new study appears in the journal Insect Molecular Biology.

"Body louse-transmitted diseases include trench fever, relapsing fever and epidemic typhus," said University of Illinois entomology professor Barry Pittendrigh, who led the research. In a previous study, Pittendrigh and his colleagues compared the sequences of all protein-coding genes in head and body lice and determined that the two belonged to the same species -- despite the fact that body lice are bigger than head lice, cling to clothing instead of hair, and can transmit disease.

Since the early 2000s, Pittendrigh has worked with John M. Clark, a professor of environmental toxicology and chemistry at the University of Massachusetts, on the molecular biology and genomics of lice. Clark was a collaborator on the 2012 study, and the two have had "a long-term goal of trying to solve this question of why body lice transmit bacterial diseases and head lice don't," Pittendrigh said.

In the new study, Clark's group infected head and body lice with Bartonella quintana, the bacterium that causes trench fever. Pittendrigh's laboratory then looked at gene expression in each to see how the insects responded to the infection.

"Our experiments suggest that the head louse immune system is fairly effective in fighting off the bacteria that cause trench fever," Pittendrigh said. "However, the body lice don't seem to have as good an immune response."

The researchers discovered that several immune genes were regulated differently in head and body lice after infection with the bacteria, and the infection progressed further in body lice over time.

"By eight days post-infection, head lice had killed or contained the invading B. quintana, whereas the bacteria were still proliferating and spreading in body lice," the researchers reported.

The team cannot yet say why head and body louse immune responses differ from one another, but Pittendrigh hypothesizes that the body louse has a reason to be more tolerant of bacterial infection.

"Head and body lice have beneficial bacteria living inside them," he said. "These bacteria make vitamins that the lice need to grow and survive. Body lice tend to grow larger than head lice. It may be that a suppressed immune system allows body lice to grow more of the bacteria that make the vitamins they need, and they grow larger."

The body louse's dampened immune response would allow other invading bacteria, such as those that cause disease in humans, to also survive in its gut, he said.

"So body lice may grow bigger, but they also are more likely to get sick with the trench fever bacteria and pass the disease to humans," Pittendrigh said.

Research demonstrates 'guided missile' strategy to kill hidden HIV

Using mouse models that have immune systems composed of human cells, researchers led by J. Victor Garcia, PhD, found that an antibody combined with a bacterial toxin can penetrate HIV-infected cells and kill them even though standard antiretroviral therapy, also known as ART, had no effect. Killing these persistent, HIV-infected cells is a major impediment to curing patients of HIV.

"Our work provides evidence that HIV-infected cells can be tracked down and destroyed throughout the body," said Garcia, professor of medicine and senior author of the study published January 9 in the journal PloS Pathogens.

For people with HIV, ART is life-saving treatment that can reduce the amount of virus in the body to undetectable levels. But as soon as treatment is stopped, the virus begins to replicate again. This means that people with HIV must be on medications for life. For some people, therapies are not without serious side effects.

In patients on ART, the virus either remains dormant or it multiplies very slowly -- it persists, hidden, even though a cocktail of drugs is aligned against it.

Garcia's findings advance the so-called "kick-and-kill" strategy for HIV eradication -- if the persistent virus is exposed, it can be targeted and killed with a new therapy.

To attack persistent HIV-infected cells, Garcia and colleagues used humanized bone marrow/liver/thymus mice -- or BLT mice -- with entire immune systems composed of human cells. This allows his team to study the distribution of persistent HIV-infected cells throughout the body and test strategies to eliminate those cells.

For the PloS Pathogens study, the researchers first treated the mice with an ART cocktail of three different drugs. Despite using strong concentrations of all three drugs, the researchers found that the virus managed to survive in immune cells in all tissues they analyzed, including the bone marrow, spleen, liver, lung, and gut.

Then they used a compound developed by co-authors Edward Berger, PhD, and Ira Pastan, PhD, from the National Institute of Allergy and Infectious Diseases (part of the National Institutes of Health). The compound is an antibody called 3B3 combined with a bacterial toxin called PE38. The researchers hypothesized that the antibody would first recognize cells expressing a specific HIV protein on the surface of infected cells. The antibody would attach to the protein and allow the toxin to enter and kill the infected cells.

When Garcia's team treated humanized HIV-infected and ART-treated mice with the 3B3-PE38 compound and then looked for infected cells in tissues, they found that the molecular missile had killed the vast majority of persistent HIV-infected cells that had been actively producing the virus despite traditional therapy, resulting in a six-fold drop in the number of infected cells throughout the immune systems.

While this reduction fell short of complete eradication, the finding offers a new route of investigation as part of the multi-pronged "kick-and-kill" strategy.

"The BLT model represents a platform in which virtually any novel approach to HIV eradication can be tested," Garcia said. "It helps us prioritize which therapeutic approaches should be advanced to clinical implementation in humans. This study shows that it's possible to attack and kill hidden HIV-infected cells that standard therapy can't touch."

It appears to be innate and stems from an evolutionary need to establish order and hierarchy in society, said San Francisco State University Professor of Psychology David Matsumoto, who co-authored the study with Hyisung Hwang, an adjunct faculty member in psychology at SF State.

In a separate study published in November, Hwang and Matsumoto also found that an athlete's culture affects the intensity with which he or she displays this body language.

"Cultures that are more status oriented have individuals who produce these behaviors more than individuals who come from cultures that are more egalitarian," said Matsumoto.

In Matsumoto and Hwang's previous research, observers labeled the body language of athletes seen in victorious poses as "triumph" and established triumph as potentially being a separate expression from pride, which requires more cognitive thinking and reflection. The new study, however, is the first to ask whether expressions of triumph are the immediate reaction of an athlete following victory.

To answer that question, Hwang and Matsumoto looked at the first body motion made by an athlete upon learning he or she was victorious, determined whether that action was among those considered to constitute "triumph," and rated the intensity of the action on a five-point scale. Actions considered triumphant included raising the arms above the shoulders, pushing the chest out, tilting the head back and smiling. They were observed in winning athletes from all cultural backgrounds and even in blind Paralympic athletes, suggesting the behavior is biologically innate.

"It is a very quick, immediate, universal expression that is produced by many different people, in many cultures, immediately after winning their combat," Matsumoto said. "Many animals seem to have a dominant threat display that involves making their body look larger."

In their other study, Hwang and Matsumoto compared the intensity of an athlete's expressions of triumph with his or her culture's "power distance" (PD), a measurement that represents the degree to which a culture encourages or discourages power, status and hierarchical differences among groups. They found that athletes from cultures with high PD produced such body language more than those from cultures with low PD.

Countries with high PD include Malaysia, Slovakia and Romania, while countries with low PD include Israel, Austria and Finland. The United States and United Kingdom fall in the middle of the PD spectrum, along with countries such as Hungary, Iran and Italy.

The results make sense, Matsumoto said, given the importance of displaying dominance to establishing status and hierarchy within a group so that the group operates efficiently. Countries that place a greater emphasis on hierarchy have a greater need for body language that helps establish power and status. But such actions can be seen in many different types of groups, he added.

"If you're in a meeting, the person sitting in the 'power chair' is going to be more erect and look taller, they're going to use a strong voice, they're going to use hand gestures that signify dominance," he said. "If there's conflict, the person who yells the most or is the most stern will be seen as the leader. It establishes the hierarchy in that context."

Additional research is needed to see if the results can be replicated across other contexts, Matsumoto said. He is also hoping to further study when these types of behaviors occur and what triggers them, as well as collect additional data to bolster the theory that triumph is a separate expression from pride.

Living on islands makes animals tamer

Jan. 10, 2014 — Most of us have seen pictures and probably YouTube videos of "tame" animals on the Galapagos Islands, the biological paradise that was Charles Darwin's major source of inspiration as he observed nature and gradually developed his ideas about the importance of natural selection as a mechanism by which populations of organisms would change -- evolve genetically -- across generations, eventually becoming better and better suited to life in their current conditions.

A corollary of Darwin's revolutionary idea was that organisms would also evolve to lose structures, functions, and behaviors they no longer needed when environmental circumstances changed. He noted that island animals often acted tame, and presumed that they had evolved to be so after coming to inhabit islands that lacked most predators.

But more than 150 years later that almost casual observation remained to come under scientific scrutiny. Today, a team of researchers from the University of California, Riverside, Indiana University Purdue University Fort Wayne and George Washington University published a study showing that island lizards are indeed "tame" as compared with their mainland relatives. The researchers were able to approach island lizards more closely than they could approach mainland lizards.

"Our study confirms Darwin's observations and numerous anecdotal reports of island tameness," said Theodore Garland, a professor of biology at UC Riverside and one of the paper's coauthors. "His insights have once again proven to be correct, and remain an important source of inspiration for present-day biologists."

Study results appear online in the Proceedings of the Royal Society B. They will appear in the journal in print on Feb. 22.

The researchers conducted analyses of relationships of flight initiation distance (the predator-prey distance when the prey starts to flee) to distance to mainland, island area, and occupation of an island for 66 lizard species, taking into account differences in prey size and predator approach speed. They analyzed island and mainland lizard species from five continents and islands in the Atlantic and Pacific Oceans and the Caribbean and Mediterranean Seas.

Their results showed that island tameness exists and that flight initiation distance decreases as distance from mainland increases. In other words, island lizards were more accessible the farther the islands were from the mainland.

"The suggestion by Darwin and others that prey on oceanic islands have diminished escape behavior is supported for lizards, which are distributed widely on both continents and islands," Garland said.

He explained that escape responses are reduced on remote islands, because predators are scarce or absent there, and natural selection under reduced predation favors prey that do not waste time and energy developing and performing needless escape.

The research team also found that prey size is an important factor that affects escape behavior.

"When prey are very small relative to predators, predators do not attack isolated individual prey," Garland said. "This results in the absence of fleeing or very short flight initiation distance."

The researchers found no conclusive evidence showing that flight initiation distance is related to island area. They found, however, that predator approach speed is an important factor in lizards.

"It is possible that other factors favor island tameness. For example, if food is scarce on islands, the cost of leaving food to flee would favor shortened flight initiation distance," Garland said.

Garland was joined in the study by William E. Cooper Jr. (first author of the research paper) at Indiana University Purdue University Fort Wayne, Ind., and R. Alexander Pyron at the George Washington University, Wash. D.C.

The study was supported in part by a grant from the National Science Foundation.

Metal ink could ease way toward flexible electronic books, displays

Wenjun Dong, Ge Wang and colleagues note that the tantalizing possibilities of flexible electronics, from tablets that roll up to wearable circuits woven into clothes, have attracted a lot of attention in the past decade. But much of the progress toward this coming wave of futuristic products has entailed making circuits using complicated, time-consuming and expensive processes, which would hinder their widespread use. In response, researchers have been working toward a versatile conductive ink. They have tried several materials such as polymers and gold and silver nanostructures. So far, these materials have fallen short in one way or another. So, Dong and Wang's group decided to try copper nanosheets, which are inexpensive and highly conductive, as a flexible circuit ink.

They made copper nanosheets coated with silver nanoparticles in the laboratory and incorporated this material into an ink pen, using it to draw patterns of lines, words and even flowers on regular printer paper. Then, to show that the ink could conduct electricity, the scientists studded the drawings with small LED lights that lit up when the circuit was connected to a battery. To test the ink's flexibility, they folded the papers 1,000 times, even crumpling them up, and showed that the ink maintained 80 to 90 percent of its conductivity.

TSMC had Q4 sales of $4.86 billion down 10% from Q3. Annual revenues were $19.88 billion – a rise of 17.8% on 2012.

December sales saw a sharp rise over November at $1.65 billion -12% up on November’s $1.47 billion which were a 14.4% drop from October’s $1.76 billion.

TSMC’s October revenues were down 6.5% on its September revenues of $1.88 billion.

TSMC had revenues of $1.86 billion in August up 5.7% on July’s $1.74 billion, but down from the $1.77 billion of June. May sales were $1.73 billion – a slight rise on April’s $1.7 billion.

TSMC’s shares dropped neatly 3% before the results announcement on rumours that Chairman Morris Chang had died. Chang then appeared at a meeting in Taiwan to scotch the rumours.

UMC had Q4 2013 sales of $1 billion, down 8% from Q3, and annual revenues of $4.1billion.

UMC had November revenues of $350 million and October revenues of $355 million.

UMC had September revenues of $340 million, August revenues of $371 million, July revenues of $386 million, June revenues of $368 million, May revenues of $361 million and April revenues of $336 million.

UMC’s 2013 annual revenue of $4.1 billion compares to $3.53 billion in 2012.
