New microscopes reveal live, developing cells in unprecedented 3-D clarity

The first microscope allows researchers to obtain fast moving images at double the spatial resolution of a conventional microscope. This provides a vastly clearer picture, enabling cell components that were once quite blurry to now become sharply defined; the difference is similar to that of a 1990's-era standard TV set versus today's high-definition TVs. The microscope is also 10 to 100 times faster than traditional technologies.

"It's always helpful to look at smaller and smaller things," said Hari Shroff, Ph.D., at NIH's National Institute of Biomedical Imaging and Bioengineering (NIBIB) lab chief of NIBIB's section on High Resolution Optical Imaging (HROI.) "Looking at a fixed cell at high resolution can tell you where different parts of the cell are at any given moment; but because much of biology depends on the movement of very small proteins finding each other and interacting, we really needed to look at how things move in a live cell."

The problem is that the higher the resolution, the harder it is to eliminate the blur from both light diffraction (the glow that sometimes occurs as light bends around objects) and the motion going on inside the live cell. Traditional linear structured illumination microscopy (SIM) cannot maintain the high resolution desired by researchers when the sample is moving quickly.

Shroff and his research fellow Andrew York, Ph.D., found an answer to these problems with their new instant linear structured illumination microscopy (iSIM), described in a paper published in Nature Methods on October 6th. Building on traditional SIM technology, the iSIM allows real-time, 3-D super resolution imaging of small, rapidly moving structures -- such as individual blood cells moving through a live zebrafish embryo. This kind of imaging is impossible with other microscopes; the ones that are fast enough to record rapid movement do not have a high enough resolution to see inside the cells; and other microscopes with similar resolution are just too slow to capture that amount of motion clearly.

If a photographer wants to take a better photograph, he can either buy a camera with a better lens and higher pixels or he can modify the picture after it's taken, using Photoshop. The principle is similar in microscopy. Instead of approaching the problem by creating better imaging software that helps to increase the resolution after the fact, as most high resolution microscopes do, Shroff and his lab developed a microscope with better lenses and mirrors so that the higher resolution is captured in the original image.

"What we've essentially done is eliminate the need for extensive computer processing by creating a better microscope at every stage of data gathering," said Shroff. "Before, we relied on computer software and algorithms to do things like sort through hundreds of images, eliminate out of focus light, and combine the individual images together. Now, we can do most of that optically with the microscope itself." This means that researchers can skip the time-consuming steps in which computers process the massive amounts of data normally required for such high resolution imaging. Now they will be able to see the images instantly instead of waiting hours or sometimes days, and the data itself takes about 1% of the hard drive space as that produced by previous microscopes.

The second microscope, described in a paper published in Nature Biotechnology online on October 13, builds on selective plane illumination microscopy (SPIM). Traditional microscopes expose the whole sample to light even though they are only imaging one small section at a time. However, just as the sun can damage skin cells, too much light exposure can damage or even kill biological samples like embryos. SPIM uses a thin beam of light to illuminate only the single plane that is currently being imaged so the biological sample is not damaged by overexposure to light. However, the technology is limited because looking at a 3-D object from only one point of view does not provide a complete representation of the object -- in the same way that viewing a globe from one perspective gives no information about what is on the other side of the world. Traditionally, SPIM microscopes rotate the sample so that they can clearly see all the dimensions, but this severely limits the imaging speed and can increase the damage done to the cells from light exposure because of the many extra images taken at multiple angles. As a result imaging is also slowed down, and the ability to capture much of the fast moving cellular motion is lost.

In order to combat this problem, Shroff and NIBIB staff scientist Yicong, Wu, Ph.D., developed a dual-view SPIM (diSPIM) microscope with two separate detection cameras. The cameras are set at a 90 degree angle to capture perpendicular views of the sample. This perpendicular view results in undistorted 3-dimensional images, and since only two views are acquired, the microscope can still capture events at very high speed. Additionally, with relatively simple modifications, traditional single camera SPIM microscopes can be converted into the dual-camera diSPIM. The real challenge in developing this technology was to find a way to combine the two disparate images from the two cameras, which required the creation of a new post-processing software algorithm.

The increased speed at which the new dual microscope can image the cells allows for clearer images of even very fast moving viruses. Being able to see how a virus enters a cell, and once it's in, how it moves around, could go a long way towards scientists' understanding of how infections occur and potentially how to fight them more effectively. In the same way, observing the migration of cancer cells in a 3-D environment could unlock information on how cancer grows, finds nutrients, and spreads.

"Biology is three-dimensional, not two dimensional. The nucleus of a cell is spherical, not circular, and as scientists, it's up to us to find ways to observe cells as accurately as possible, Shroff said. "We're really moving biology into the third dimension with this microscope." There's a lot of attention right now on how neurons fire and interact with each other, but the truth is, we don't even understand how a brain develops -- even in the most simple of organisms like C. elegans, a worm with only 300 brain cells. We don't know why brain cells go where they do or what determines their organization. We can't understand more about this process without observing it, and that's something that these devices can help to provide."

The Shroff lab has already begun multiple collaborations with biological labs both inside the NIH as well as external institutions, including Yale, Sloan Kettering, and the University of Connecticut Health Center.

UntitledThe UK Electronics Skills Foundation has announced industry backing for 60 scholarships for the 2013/14 academic year.

This takes promised industrial support for the electronic engineering scholarship programme to over £1m in three years.

It has helped 180 engineering students through university and into rewarding electronic engineering careers.

The scholarships are available to students studying electronic engineering at any of the 11 UKESF partner universities, all leading UK universities recognised for their strength in electronics teaching and research.

This is intended to help address the shortage of skilled students entering the electronics industry in the UK, an industry which the government now recognises it important for economic growth.

This year’s Electronic Systems industry report for government, ESCO, stated innovation in the sector is an enabling driver in virtually all segments of the UK economy, contributing £78 billion ($125bn) and employing 850,000 skilled workers.

“The UK electronics sector is a vital yet fragmented industry so we must work collectively to promote awareness of its significance, global impact and wide ranging career opportunities,” said UKESF chairman and Plastic Logic CEO Indro Mukerjee.

According to UKESF programme director, Wendy Daniell: “UCAS data show a higher proportion of students are now applying for vocational degrees, perhaps as a result of rising tuition fees. Student degree choices are increasingly being influenced by factors such as graduate employment prospects and scholarships.”

UntitledFisnar Europe is taking its latest multi capability gasket -forming and PCB encapsulation system to Munich next month.

This is the first time that partner companies Fisnar Europe and Liquid Control have attended the Productronica exhibition in Munich.

“The F9960N is a strong addition to the Fisnar Liquid Control product portfolio and we are excited to be launching it at Productronica,” said company managing director, John Henderson.

The F9960N Robot system has been designed for materials dispensing applications ranging from miniature SMT circuit boards to larger PCB’s and finished assemblies.

It will be used for precision form-in-place gaskets, encapsulation, coating, EMI and thermal-grease applications as well as underfill and sealing projects.

It is the first Fisnar gantry robot to be mounted within an enclosed free-standing platform.

“The enclosed environment provides access for fume extraction systems, safeguarding the work force and work place from potentially hazardous substances,” said the company.

Control is via an interactive 178mm touch- screen display supplied with the system. Other features include software nozzle/tip change alignment routine for quick and automatic program change offsets and step-and-repeat functions for quick programming of multiple and identical objects.

Nordson Yestech will show its FX SL advanced AOI inspection system for PCBs at Productronica exhibition in Munich ( November 12 – 15).

UntitledThe FX SL AOI, which will be demonstrated on the Yestech Stand 438 in Hall A2, offers high speed “capture on the fly” inspection using angled cameras, a 3D height sensor, full color digital image processing technology and both image- and rule-based algorithms.

Configurable for all line positions, the system can be used for paste, pre / post-reflow and final assembly inspection.

The use of advance colour imaging offers “high-speed PCB inspection with exceptional defect coverage”, said the supplier.

With one top-down viewing camera and four side-viewing cameras, the FX SL inspects solder joints and verifies correct part assembly.

Fungus that causes white-nose syndrome in bats proves hardy survivor

Their report appears in the journal PLOS ONE.

The aptly named fungus Pseudogymnoascus (Geomyces) destructans causes white-nose syndrome in bats. The infection strikes bats during their winter hibernation, leaving them weakened and susceptible to starvation and secondary infections. The fungus, believed to have originated in Europe, was first seen in New York in the winter of 2006-2007, and now afflicts bats in more than two dozen states. According to the U.S. Fish and Wildlife Service, P. destructans has killed more than 5.5 million bats in the U.S. and Canada.

The fungus thrives at low temperatures, and spreads to bats whose body temperature drops below 20 degrees Celsius (68 degrees Fahrenheit) when they are hibernating in infected caves. Previous research has shown that the fungus persists in caves even after the bats are gone.

The new study, from researchers at the Illinois Natural History Survey at the University of Illinois, found that the fungus can make a meal out of just about any carbon source likely to be found in caves, said graduate student Daniel Raudabaugh, who led the research under the direction of survey mycologist Andrew Miller.

"It can basically live on any complex carbon source, which encompasses insects, undigested insect parts in guano, wood, dead fungi and cave fish," Raudabaugh said. "We looked at all the different nitrogen sources and found that basically it can grow on all of them. It can grow over a very wide range of pH; it doesn't have trouble in any pH unless it's extremely acidic."

"P. destructans appears to create an environment that should degrade the structure of keratin, the main protein in skin," Raudabaugh said. It has enzymes that break down urea and proteins that produce a highly alkaline environment that could burn the skin, he said. Infected bats often have holes in their skin, which can increase their susceptibility to other infections.

The fungus can subsist on other proteins and lipids on the bats' skin, as well as glandular secretions, the researchers said.

"P. destructans can tolerate naturally occurring inhibitory sulfur compounds, and elevated levels of calcium have no effect on fungal growth," Raudabaugh said.

The only significant limitation of the fungus besides temperatures above 20 degrees Celsius has to do with its ability to take up water, Raudabaugh said. Its cells are leaky, making it hard for the fungus to absorb water from surfaces, such as dry wood, that have a tendency to cling to moisture. But in the presence of degraded fats or free fatty acids, like those found on the skin of living or dead animals, the fungus can draw up water more easily, he said.

"All in all the news for hibernating bats in the U.S. is pretty grim," Miller said.

"When the fungus first showed up here in Illinois earlier this year we went from zero to 80 percent coverage in a little more than a month," he said. The team led by U. of I. researchers that discovered the fungus in the state found a single infected bat in one northern Illinois cave, he said. Several weeks later most of the bats in that cave were infected.

Although many studies have been done on the fungal genome and on the bats, Miller said, Raudabaugh is the first to take an in-depth look at the basic biology of the fungus.

"Dan found that P. destructans can live perfectly happily off the remains of most organisms that co-inhabit the caves with the bats," Miller said. "This means that whether the bats are there or not, it's going to be in the caves for a very long time."

The Illinois Natural History Survey is a division of the Prairie Research Institute at the U. of I.

Typical power consumption is approximately 100mW for composite inputs, said the firm, and a power-down mode further reduces consumption.

Called TW9990, the chip supports NTSC, PAL, SECAM, single-ended, differential, and pseudo differential composite video inputs as well as S-Video.

Direct support for differential inputs eliminates the need for an external op-amp, and a

single 27MHz crystal works for all the video standards.

IT also has built-in programmable short-to-battery and short-to-ground detection. “This is an essential functionality required by OEMs when supporting a differential signal input,” claimed Intersil.

Use is expected in automotive rear camera displays.

Grasshopper mice are numb to the pain of the bark scorpion sting

The grasshopper mice are essentially numb to the pain, scientists have found, because the scorpion toxin acts as an analgesic rather than a pain stimulant.

The scientists published their research this week in Science.

Ashlee Rowe, lead author of the paper, previously discovered that grasshopper mice, which are native to the southwestern United States, are generally resistant to the bark scorpion toxin, which can kill other animals.

It is still unknown why the toxin is not lethal to the mice.

"This venom kills other mammals of similar size," said Rowe, Michigan State University assistant professor of neuroscience and zoology. "The grasshopper mouse has developed the evolutionary equivalent of martial arts to use the scorpions' greatest strength against them."

Rowe, who conducted the research while at The University of Texas at Austin, and her colleagues ventured into the desert and collected scorpions and mice for their experiments.

To test whether the grasshopper mice felt pain from the toxin, the scientists injected small amounts of scorpion venom or nontoxic saline solution in the mice's paws. Surprisingly, the mice licked their paws (a typical toxin response) much less when injected with the scorpion toxin than when injected with a nontoxic saline solution.

"This seemed completely ridiculous," said Harold Zakon, professor of neuroscience at The University of Texas at Austin. "One would think that the venom would at least cause a little more pain than the saline solution. This would mean that perhaps the toxin plays a role as an analgesic. This seemed very far out, but we wanted to test it anyway."

Rowe and Zakon discovered that the bark scorpion toxin acts as an analgesic by binding to sodium channels in the mouse pain neurons, and this blocks the neuron from firing a pain signal to the brain.

Pain neurons have a couple of different sodium channels, called 1.7 and 1.8, and research has shown that when toxins bind to 1.7 channels, the channels open, sodium flows in and the pain neuron fires.

By sequencing the genes for both the 1.7 and 1.8 sodium channels, the scientists discovered that channel 1.8 in the grasshopper mice has amino acids different from mammals that are sensitive to bark scorpion stings, such as house mice, rats and humans. They then found that the scorpion toxin binds to one of these amino acids to block the activation of channel 1.8 and thus inhibit the pain response.

"Incredibly, there is one amino acid substitution that can totally alter the behavior of the toxin and block the channel," said Zakon.

The riddle hasn't been completely solved just yet, though, Rowe said.

"We know the region of the channel where this is taking place and the amino acids involved," she said. "But there's something else that's playing a role, and that's what I'm focusing on next."

Some resistance to prey toxins in mammals has been found in other species. The mongoose, for example, is resistant to the cobra. And naked mole rats' eyes do not burn in pain when carbon dioxide builds up in their underground tunnels.

This study, however, is the first to find that an amino acid substitution in sodium channel 1.8 can have an analgesic effect.

Rowe said studies such as this could someday help researchers target these sodium channels for the development of analgesic medications for humans.

Although the importance of stem cells in fuelling plant growth and development still many questions on their tight molecular control remain unanswered. Plant researchers at VIB and Ghent University discovered a new step in the complex regulation of stem cells.

Lieven De Veylder said, "Our data suggest that certain organizing stem cells in plant roots are less sensitive for DNA-damage. Those cells hold an original and intact DNA copy which can be used to replace damaged cells if necessary. Animals rely on a similar mechanism but most likely plants have employed this in a more optimized manner. This could explain why many plants can live for more than hundreds of years, while this is quite exceptional for animals."

Quiescent organisers of plant growth

Plant growth and development depend on the continuous generation of new cells. A small group of specialized cells present in the growth axes of a plant is driving this. These so-called stem cells divide at a high frequency and have the unique characteristic that the original mother cell keeps the stem cell activity while the daughter cell acquires a certain specialization. Besides these stem cells, plant roots also harbor organizing cells. These organizing cells divide with a three- to ten-fold lower frequency, therefore often referred to as quiescent center cells. The organizing cells control the action of the surrounding stem cells and can replace them if necessary.

A new molecular network

For almost 20 years, scientists all over the world have been studying the action of the stem cells and that of their controlling organizing cells. Until now it was not known how quiescent and actively dividing cells could co-exist so closely and which mechanisms are at the basis of the quiescent character. Plant researchers at VIB and Ghent University have now identified a new molecular network that increases our understanding of stem cell regulation and activity.

Central in this process is the discovery of a new protein, the ERF115 transcription factor. The scientists demonstrated that the organizing cells barely divide because of the inhibition of ERF115 activity. When the organizing cells need to divide to replace damaged surrounding stem cells, ERF115 gets activated. ERF115 then stimulates the production of the plant hormone phytosulfokine which in turn activates the division of the organizing cells. Thus, the ERF115-phytosulfokine network acts as a back-up system during stress conditions which are detrimental for the activity of stem cells.

AtmelMaxTouchAtmel has announced another generation of capacitive screen touch controllers, claiming them to be significantly better at dealing with big fingers, small styluses, water and gloved fingers.

Compared with its last generation, there are now 20 DACs taking data off 20 sense wires followed by parallel processing, compared with 18 wires multiplexed onto fewer ADCs before.

Having moved to a 130nm process for the new chips, there was also room to replace the old 8bit processor with a 32bit AVR core.

“It is a lot more power efficient and has a lot more processing capability for a lot more intelligence: large finger detection, palm rejection, water rejection and charger noise immunity” Atmel product marketing manager Tony Xia told Electronics Weekly. “And it can work with xSense, our scheme to use fine copper wires to replace ITO.”

As well as replacing increasingly hard to get ITO (indium tim oxide), xSense is claimed to reduce sense line impedance which allows the wire to be charged and read faster. With more sense cycles in a fixed time, statistical processing can reduce the effective signal-to-noise ratio.

Charger noise rejection, said Xia, is actually a combination of better numerical processing, actively hopping the read frequency away from the charger band, and analogue processing.

The new flagship device is the 642T offering 32×20 conductors, compared with 30×18 with former top model 540F.

On water, Xai said: “Condensation with a finger, there is no impairement; wet hands from washing, there is no problem; up to a few drops, there is no false touch; and it automatically works after wiping.”

And on gloves: “A lot of smartphones today won’t work with gloves at all. Ours worked well with 20 different gloves we were presented with on one customer visit.“

And the problem with big fingers is, according to Xia, that they can be falsely detected as multiple small fingers grouped together.

Lastly, passive styluses no longer need to be conductive and in electrical contact with the user. “We tried it with a cheap mechanial pencil with an 0.5mm lead and it worked,” said Xia

Increasing toxicity of algal blooms tied to nutrient enrichment and climate change

Oct. 24, 2013 — Nutrient enrichment and climate change are posing yet another concern of growing importance: an apparent increase in the toxicity of some algal blooms in freshwater lakes and estuaries around the world, which threatens aquatic organisms, ecosystem health and human drinking water safety.

As this nutrient enrichment, or "eutrophication" increases, so will the proportion of toxin-producing strains of cyanobacteria in harmful algal blooms, scientists said.

Researchers from Oregon State University and the University of North Carolina at Chapel Hill will outline recent findings in an analysis Friday in the journal Science.

Cyanobacteria are some of the oldest microorganisms on Earth, dating back about 3.5 billion years to a time when the planet was void of oxygen and barren of most life. These bacteria are believed to have produced the oxygen that paved the way for terrestrial life to evolve. They are highly adaptive and persistent, researchers say, and today are once again adapting to new conditions in a way that threatens some of the life they originally made possible.

A particular concern is Microcystis sp., a near-ubiquitous cyanobacterium that thrives in warm, nutrient-rich and stagnant waters around the world. Like many cyanobacteria, it can regulate its position in the water column, and often forms green, paint-like scums near the surface.

In a high-light, oxidizing environment, microcystin-producing cyanobacteria have a survival advantage over other forms of cyanobacteria that are not toxic. Over time, they can displace the nontoxic strains, resulting in blooms that are increasingly toxic.

"Cyanobacteria are basically the cockroaches of the aquatic world, they're the uninvited guest that just won't leave," said Timothy Otten, a postdoctoral scholar in the OSU College of Science and College of Agricultural Sciences. His work has been supported by the National Science Foundation.

"When one considers their evolutionary history and the fact that they've persisted even through ice ages and asteroid strikes, it's not surprising they're extremely difficult to remove once they've taken hold in a lake," he said. "For the most part, the best we can do is to try to minimize the conditions that favor their proliferation."

Researchers lack an extensive historical record of bloom events and their associated toxicities to put current observations into a long-term context. However, Otten said, "If you go looking for toxin-producing cyanobacteria, chances are you won't have to look very long until you find some."

There are more than 123,000 lakes greater than 10 acres in size spread across the United States, and based on the last EPA National Lakes Assessment, at least one-third may contain toxin-producing cyanobacteria. Dams; rising temperatures and carbon dioxide concentrations; droughts; and increased runoff of nutrients from urban and agricultural lands are all compounding the problem.

Many large, eutrophic lakes such as Lake Erie are plagued each year by algal blooms so massive that they are visible from outer space.

Researchers studying cyanobacterial toxins say it's improbable that their true function was to be toxic, since they actually predate any predators. New research suggests that the potent liver toxin and possible carcinogen, microcystin, has a protective role in cyanobacteria and helps them respond to oxidative stress. This is probably one of the reasons the genes involved in its biosynthesis are so widespread across cyanobacteria and have been retained over millions of years.

Because of their buoyancy and the location of toxins primarily within the cell, exposure risks are greatest near the water's surface, which raises concerns for swimming, boating and other recreational uses.

Also, since cyanobacteria blooms become entrenched and usually occur every summer in impacted systems, chronic exposure to drinking water containing these compounds is an important concern that needs more attention, Otten said.

"Water quality managers have a toolbox of options to mitigate cyanobacteria toxicity issues, assuming they are aware of the problem and compelled to act," Otten said. "But there are no formal regulations in place on how to respond to bloom events.

"We need to increase public awareness of these issues," he said. "With a warming climate, rising carbon dioxide levels, dams on more rivers than not, and overloading of nutrients into our waterways, the magnitude and duration of toxic cyanobacterial blooms is only going to get worse."
